How we can auto create single PO from multiple Purchase Requisition? Any idea or suggestion ?

If all the requisitions have some common criteria in them (e.g. same requestor or same supplier) then simply query them up from Auto create window and convert them to a PO.

When you create PO from autocreate, from Action, select Add To.
